Please fill out the form to the left to submit a request for Cell Sorter usage. Core staff will follow the workflow and policies below with regards to reservation confirmation for both sorting and analysis.

If you need an account created on any of our analyzer instruments, or if you need a template moved from one instrument to another, please fill out the form to the left and the core staff will make that account. COVID Workflow
For Cell Sorting, the workflow and guidelines are as follows:

For External Cell Sorting, the workflow and guidelines are as follows for the early phase return to research. Dana Farber Cancer Institute has granted access to Brigham and Women’s Hospital and Children’s hospital researchers to the DFCI Flow Cytometry Core as of 10-1-20. Please note this permission allows you access to the sorting facility only, there is no external researcher access to the analyzers.

External Cell Sorting Guidelines
- Scheduling is no longer performed by the researcher in iLabs.
- All scheduling requests must be done via the website Sorting Reservation Request Form. http://flowcytometry.dana-farber.org/schedule.html.
- Researchers are required to provide a valid Purchase Order when scheduling.
- PO must be entered exactly as it appears on the PO you provided. If it is incorrect or missing digits your reservation request will be deleted.
- For a new PO, a PDF copy of the PO must be emailed to [email protected]u
- The PDF must have the correct shipping address for Dana Farber Cancer Institute, date, and amount.

Cell Sorting Guidelines
- NEW!!! The core requires an extra scheduled 30 minutes per sort.
15 minutes at the beginning of the sort and your sort will end 15 minutes before the end of the reservation - Place container in bin outside sorter facility on Mayer 3
- Sorting will be scheduled up to three weeks in advance
- Samples must be capped, and collection tubes must be brought in a container
- Sort samples should be placed in containment bins outside sorter facility on Mayer 3
- Samples should be named numerically only. We want to avoid any errors
- Scheduling requests will be still be submitted via smart sheet
- Staff will review request and contact researcher directly
- Staff will be inviting the researcher to a recorded zoom meeting to review the cells, sorting parameters and gating populations
- Sample filtration and re-suspension will be done by core staff if needed
- In the case of problematic samples, we will be in contact via cell phone or Zoom, so users should plan to be available for contact during their reservation
- If staff cannot contact you, they will use best judgment to prioritize and troubleshoot issues within the confines of the reservation time, extra time will not be offered if unavailable
- All cancellation policies here are still in effect

General Policies
- Last researcher of each day is responsible for filling the Sheath fluid
- Anyone using the instrument after 5pm weekdays or anytime during the weekends is required to empty the waste after each reservation
- Only one user per active instrument is permitted (NO EXCEPTIONS)
- Under no circumstances should you be in one of these rooms if you are not running an experiment.
- Only trained researchers should be analyzing experiments
- There will be virtual training of new researchers at this time due to the DFCI social distancing policy
- Researchers are not allowed to train lab mates. The Flow Core are responsible for all training
- The core cannot physically assist with experiments. Zoom assistance can be scheduled in advance
- In the case of problematic instrumentation, please call 617-632-3179 or email [email protected] immediately
- All cancellation policies here are still in effect
Cleaning Policies
- Cleaning post analysis is required to use a virucidal wipe on:
- Fortessa buttons (run, flow rates, standby and prime buttons)
- Desk, keyboard and mouse
- Biohazard waste carry in / carry out waste will be strictly enforced
- Bring small bio-hazard bag with you
- Absolutely no waste should be thrown away in the regular trash
Diva
- We are requiring all researchers to establish a password in DIVA.
- This will ensure the person who is logged into Diva is the correct person being invoiced. Do not share accounts.
HTS plate mode
- You must return the instrument to tube mode when finished.
- If you are analyzing plates you must stop 10min prior to the end of your
reservation to run the cleaning plate (8min)
- DO NOT IMPORT experiments containing DATA back on the computer
- DO NOT ADD DATA to an experiment from a previous acquisition
- DO NOT ADD PLATES to an experiment from a previous acquisition run
- DO NOT EXPORT an EXPERIMENT TEMPLATE
- If the experiment already contains DATA
- Copy and paste the experiment so it contains no data
- Delete all tubes except the last tube
- THEN EXPORT experiment template
- DO NOT EXPORT an EXPERIMENT TEMPLATE
- If the experiment already contains PLATE DATA
- DO NOT USE THE "Duplicate Without Data" function
- Copy your experiment and paste with no data
- DO NOT Allow your experiment to exceed 500MB of data
- Option 1. Stop acquiring Data > Import the template and continue in the new experiment
- Option 2. Stop acquiring Data > Copy Experiment > Paste on your directory name (top of Browser window)
- Experiments over 500MB will not be stored on the core server
- Experiments should be named with an identifying label. If this is not done, experiments may overwrite others during core export
